Hypericum perforatum L. plant, which is widely spread in our country, is used in the treatment of many diseases such as colds, depression, gallbladder diseases, bladder irritations, migraine pains and burns. In this study, it was aimed to determine the tyrosinase and collagenase inhibitory potentials of two different St. John’s wort oils obtained from a pharmacy (sample 1) and a herbalist (sample 2). Experiments were carried out on enzyme reactions based on colorimetric measurement. Both samples were studied at a concentration of 20 µg/mL in all experiments. It was determined that sample 1 (18.3±0.15% inhibition) provided higher collagenase enzyme inhibition compared to sample 2 (4.1±0.28% inhibition) and again sample 1 (15.1±0.24% inhibition) provided higher tyrosinase enzyme inhibition compared to sample 2 (3.7±0.19% inhibition). According to these results; it was determined that St. John’s Wort oil inhibits collagenase and tyrosinase enzymes in general, and that St. John’s Wort oil from pharmacies provides higher inhibition than St. John’s Wort oil from herbalists. It can be interpreted that this situation occurs as a result of the producer collecting the plant at the right time, drying it appropriately and producing oil with the right maceration method; thus, the density of the bioactive components in it may have increased, contributing to the increase in activity.
