Molecular Identification of Globodera Species in Potato Cyst Nematode Populations in Niğde Province

Molecular identification was performed on Potato Cyst Nematode (PCN) populations taken from 38 locations where intensive potato cultivation is carried out in Niğde province. Globodera rostochiensis, named as Golden Cyst Nematode, and Globodera pallida, named as White Cyst Nematode, are species that cause serious yield losses in potato cultivation in our country and worldwide. G. rostochiensis, which was first detected in Bolu province in Türkiye in 1985, was also detected in potato fields in the Aegean Region in the following years. Accurate and rapid species identification is very important in resistant variety development studies, which is the most effective control strategy against PCN. G. rostochiensis and G. pallida were identified using molecular techniques. The ITS5, PITSp4, and PITSr3 primers were employed to differentiate between these nematode species with precision. As a result of species identification, all samples were identified as G. rostochiensis.
