Investigation of the Complementary Therapeutic Potentials of Some Phytotherapeutic Plants on Breast Cancer Cell Lines

Today, the use of herbal therapy methods is increasingly preferred to enhance the therapeutic effectiveness and reduce the side effects of drugs synthesized with modern techniques. Extracts obtained from many plants have antimicrobial, anti-inflammatory, anti-cancer, and other effects. However, when these plants used in combination with cancer drugs, may yield results that enhance the effectiveness of treatment. Based on this, two different plant extracts (Urtica sp and Pinus sp) were combined with Doxorubisine respectively. After obtaining the plant extracts using wet-laboratory methods, their cytotoxicological effects, cellular growth rates, and nucleus morhphological changes were examined. As a result, the IC50 value of the active ingredient Doxorubicin was determined to be 19.8 µM, and that of the plants as 330 µg/mL for Urtica sp. and 957 µg/mL for Pinus sp. respectively. At the lowest toxicity IC5 values, viability rates showed a decrease of 33.7% and 38.2% for Urtica sp and Pinus sp, respectively. The combined use of the Doxorubicin with the aforementioned plants has not been previously demonstrated on cancer cells, and their therapeutic potential was investigated for the first time within the scope of our study. According to our results, Urtica sp. and Pinus sp. extracts increased the sensitivity of breast cancer cells to Doxorubicin and caused regressions in both cellular growth and cell migration. According to the Bliss Independence model, the plant extracts with Doxorubicin showed a synergistic effect. Accordingly, significant data regarding the complementary therapeutic properties of the plants we tested were revealed, and the results obtained serve as a guide for future research on the subject.
