Rheum Ribes Ekstraktlarının Biyoaktif Bileşimi, Antioksidan ve Sitotoksik Aktiviteleri

Bu çalışmanın amacı, Rheum ribes kökü (RTE) ile kök ve genç sürgünlerin (RYSE) etanol ekstraktını kimyasal bileşimi, antioksidan özellikleri ve sitotoksik etkisini değerlendirmektir. Rheum ribes ekstraktlarının toplam polifenol (TPC), toplam flavonoid (TFC), radikal yakalama aktivitesi (DPPH), antioksidan gücü (FRAP) kolorimetrik yöntemler kullanılarak belirlendi. Örneklerin mineral içerikleri ve vitamin değerleri sırasıyla ICP-MS ve HPLC ile belirlendi. Rheum ribes ekskratlarının sitotoksik etkisi, XTT metodu kullanılarak farklı hücre hatları üzerinde belirlendi. Kanser hücrelerinde RTE ve YSE'nin sitotoksik etkileri ile apoptoz açısından değerlendirildi. Sonuçlara göre mineral içerikleri, vitamin A ve C değerleri her iki örnekte de yüksek bulundu. Rheum ribes genç sürgününün etanolik ekstraktları, WI-38 hücrelerine kıyasla tüm kanser hücreleri üzerinde seçici sitotoksik etki göstermiş ve ekstraktın kanser hücrelerinde IC50 değerleri 26.10 -54.81 μg/mL arasında bulundu. RTE ve RYSE'nin etanolik ekstrakları, MCF-7 hücrelerinde apoptozu indükledi. Rheum ribes'in etanolik ekstraktı güçlü sitotoksik ve antioksidan aktiviteye sahiptir. Kanser hücreleri üzerindeki sitotoksik etki mekanizmalarını belirlemek için daha fazla araştırmaya ihtiyaç vardır.
