Determination of Bacteria Levels Contained in Some Probiotic Preparations Consumed and Confirmation by PCR

Abstract

This study aimed to investigate the viability and levels of bacteria as indicated on the label in probiotic preparations used as supplements. Forthis purpose, a total of 15 different preparations containing LactobacillusandBifidobacterium species belonging to different brands were obtained. The sampleswere subjected to microbiological cultivation in terms of Lactobacillus andBifidobacterium species. As a result of the microbiological analysis, live bacteria were detected in 13 (%87) of the samples, while no growth was observed in 2 (%13) samples. Lactobacillus spp. average number of bacteria in probiotic preparations containing 5,9×1010 cfu/g;Bifidobacterium spp.on the other hand, in probiotic preparations containing bacteria, the average number of bacteria was 1,3×109 cfu/g. Strains that underwent Gram staining, catalase test and carbonhydrate fermentation test were confirmed by PCR analysis. As a result of PCR analysis, Lactobacillus strains and Bifidobacterium strains were confirmed in 13 samples. As a result, the presence, viability, and quantity of strains specified in ready-to-use probiotic preparations are very important. Our study showed that the accuracy of the information written on the label was 87%.
